Obesity is a worldwide health problem directly linked to several disease processes such as hypertension and type 2 diabetes melitus[1]. Adipose tissue is not only an organ for passive energy reserve, but also an active endocrine organ secreting a wide range of hormones and other protein factors called adipokines[1, 2]. Among the adipokines, resistin is involved in insulin sensitivity and glucose tolerance[3, 4] while others are involved in hemostasis, inflammatory and stress responses, and energy balance[5, 6]. Resistin, initially identified in screening for adipocytespecific transcripts down-regulated by treatment with thiozolidinedione TZDs ; , belongs to a novel family of cysteine-rich proteins, each with a unique tissue distribution[3, 4]. In rodents, resistin predominantly expressed in white adipose tissue[4] reduces insulin sensitivity in adipocytes and skeletal muscles by impairing insulin-mediated glucose transport and inducing the expression of suppressor of cytokine signaling 3 SOCS3 ; [7-9], and regulates fasting blood glucose by increasing hepatic glucose release[10]. Therefore, resistin might provide a link between obesity and diabetes melitus. Initial studies on the regulation of resistin indicate that resistin expression is reduced by fasting and increases rapidly on refeeding [3]. Decreased immune function in cancer patients is well established, 1 and it is self-evident that clinically apparent tumors avoid effective antitumor immune responses. Tumor-associated immune defects are observed in all immune organs and cell lineages, including the thymus and T cells.2-8 While thymic atrophy accompanies normal aging, 9 a high incidence of premature thymic involution is seen in childhood malignancies, 10 and this often rebounds after curative treatment. Serum size fractionation studies have shown thymic atrophy associated with a nonimmunoglobulin serum component with a molecular weight higher than 25 000 Da in mice that received transplants of Lewis lung carcinoma cells.15 Consistent with this finding, VEGF 42 kDa ; and its receptors have profound effects on the early development and differentiation of both vascular and hematopoetic progenitors, 16 and VEGF receptors are present on some of the earliest hematopoetic progenitor cells HPCs ; .17 We have previously demonstrated that VEGF, a factor produced by a large fraction of solid tumors, causes a defect in the functional maturation of dendritic cells DCs ; from early HPCs.18, 19 In addition to DC defects, nontumor-bearing mice treated with a continuous infusion of recombinant VEGF have a decreased number of T cells and a decreased T-celltoB-cell ratio in their lymph nodes and spleen, 19 and thus VEGF represents a candidate mediator of the tumor-associated thymic defect. This increased B-cell population may be compensatory in nature and suggests that in addition to causing defective developmental progression within the myeloid lineage, lymphoid lineage commitment may be similarly effected by VEGF. In this study we investigate whether pathologically relevant concentrations of VEGF inhibit T-cell development, and we explore the cellular mechanisms involved in this process.

We deal with a model scalar nonlinear convection-diffusion equation. A semi-discretization with respect to the space coordinates is carried out by the discontinuous Galerkin finite element method DGFEM ; with nonsymmetric treatment of stabilization terms and interior and boundary penalty. Such scheme was analysed in [3] for elliptic problems and we extend this analysis also for convectiondiffusion problems in [2]. The semi-discretization with respect to space leads to a system of ODE, which can be discretized by a suitable method. A use explicit scheme causes a strong restriction to the choice of the time step. A full implicit scheme leads to a necessity to solve a nonlinear system of algebraic equations in each time step. Therefore, we proposed in [1] a first-order semi-implicit method, where the linear terms are treated implicitly and the nonlinear ones explicitly. We present a generalization of this approach to a higher order approximation with respect to the time using the backward difference formulae BDF ; . The linear terms are discretized with the aid of multistep BDF and for the nonlinear ones we use an explicit higher order extrapolation which preserves the stability and accuracy of the numerical scheme. We analyse this method and prove a priori order of convergence of order O hp + -norm and H 1 -seminorm, where h and are the mesh size and time step, respectively, p is the degree of polynomial approximation of DGFEM and n is the order of BDF.
