Whether the anti-atherogenic ability of D-4F requires endogenous apoA-I is unknown. Objective: To determine if D-4F inhibits atherosclerosis in the chow-fed LDLR apobec-1 null mouse model and whether the anti-atherogenic effects of D-4F are dependent on the presence of endogenous apoA-I. Methods: LDLR apobec-1 double knockout DKO ; and LDLR apobec-1 apoA-I triple knockout TKO ; mice were administered D-4F orally in drinking water 0.5mg ml ; for 16 weeks and compared with parallel controls that received no D-4F. The extent of atherosclerosis in the aortic root was determined after staining with oil red O. Results: Oral administration of D-4F for 16 weeks significantly reduced aortic root atherosclerosis by 28% p 0.01 ; in the DKO mice and by 38% p 0.01 ; in the TKO mice. Whole serum cholesterol efflux studies using 16 week sera showed significantly increased scavenger receptor BI- SR-BI ; mediated efflux in D-4F treated mice in both the DKO and TKO groups, whereas adenosine triphosphate binding cassette A-1 ABCA1 ; -mediated cholesterol efflux was unchanged. Conclusion: Oral administration of D-4F significantly reduced atherosclerosis in chow-fed LDLR apobec-1 DKO mice. Furthermore, D-4F reduced atherosclerosis in TKO mice lacking endogenous apoA-I. D-4F administration enhanced SR-BI-dependent cholesterol efflux capacity of serum in both groups. These results demonstrate that the anti-atherogenic effects of D-4F, which may include SR-BI-dependent efflux promotion, are independent of endogenous apoA-I.

The present findings extend our knowledge regarding the cellular and molecular determinants of an enhanced 3-AR mediated negative inotropic effect in CHF, provide valuable new insight into the mechanism of the progression of functional impairment in CHF, and may assist in specifically targeting therapy for CHF and ferret.
