The mutants were analysed for HPRT deletions with PCR-based methods aiming at detection of retention, loss or change of eight gene fragments containing the exons and flanking intron sequences of genomic HPRT DNA. In the population of healthy controls who have been further analysed in the present work, ~19% of mutants were found to contain a genomic HPRT deletion 14 ; . Mutant clones, which showed `no detectable change' in the deletion analysis and contained sufficient amount of DNA for further study, were selected for analysis of point mutations by reverse transcriptase--PCR and DNA sequencing methods.
